OnabotulinumtoxinA


Pronunciation: OH-na-BOT-ue-li-num-TOX-in-A
Generic Name: OnabotulinumtoxinA
Brand Name: Botox Cosmetic

OnabotulinumtoxinA may spread from the injection site to other areas of the body, causing symptoms of a serious condition called botulism. These symptoms may occur hours to weeks after you receive OnabotulinumtoxinA. Symptoms may include loss of strength; muscle weakness; double or blurred vision; drooping eyelids; hoarseness; change or loss of voice; loss of bladder control; or trouble speaking, breathing, or swallowing. The risk may be greater in children being treated for spasticity (muscle spasms), but these symptoms may also occur in adults. OnabotulinumtoxinA is not approved for use in children. Contact your doctor immediately if these symptoms occur.


Severe and sometimes fatal breathing or swallowing problems have been reported. The risk may be greater in patients who already have breathing or swallowing problems. Tell your doctor if you have or are at risk of developing breathing or swallowing problems.





OnabotulinumtoxinA is used for:

Temporarily improving the appearance of moderate to severe lines between the eyebrows in patients 65 years old or younger. It may also be used for other conditions as determined by your doctor.


OnabotulinumtoxinA is a neurotoxin. It works by blocking nerve impulses to the muscles or sweat glands, temporarily paralyzing the muscle or gland.


Do NOT use OnabotulinumtoxinA if:


  • you are allergic to any ingredient in OnabotulinumtoxinA or another botulinum product

  • you have an infection at the injection site

  • you are pregnant

Contact your doctor or health care provider right away if any of these apply to you.



Before using OnabotulinumtoxinA:


Some medical conditions may interact with OnabotulinumtoxinA. Tell your doctor or pharmacist if you have any medical conditions, especially if any of the following apply to you:


  • if you are pregnant, planning to become pregnant, or are breast-feeding

  • if you are taking any prescription or nonprescription medicine, herbal preparation, or dietary supplement

  • if you have allergies to medicines, foods, or other substances

  • if you have nerve problems or disease (eg, amyotrophic lateral sclerosis, motor neuropathy), muscle problems or disease (eg, Lambert-Eaton syndrome, myasthenia gravis), bleeding disorders, seizures, or severe weakness or wasting of the muscles at the injection site (eg, trouble raising your eyebrows, drooping eyelids)

  • if you have a history of heart problems (eg, irregular heartbeat, a recent heart attack), swallowing or breathing problems (eg, asthma), or eye problems (eg, glaucoma, drooping eyelid)

  • if you have a skin infection; or inflammation, scarring, or other skin disorders at the injection site

  • if you plan to have surgery, especially surgery on your face, or if you have received OnabotulinumtoxinA or any other botulinum toxin within the past 4 months

  • if you have weakness in your forehead muscles, have had any change in the way your face normally looks, or you have had surgery on your face

  • if you are taking an allergy or cold medicine

Some MEDICINES MAY INTERACT with OnabotulinumtoxinA. Tell your health care provider if you are taking any other medicines, especially any of the following:


  • Aminoglycoside antibiotics (eg, gentamicin), anticholinesterase medicines (eg, neostigmine), lincosamides (eg, clindamycin), magnesium, muscle relaxants (eg, cyclobenzaprine), neuromuscular blockers (eg, atracurium), polymyxin, quinidine, or sleep medicines (eg, zolpidem) because they may increase the risk of OnabotulinumtoxinA's side effects

  • Anticoagulants (eg, warfarin) because the risk of bleeding at the injection site may be increased

How to use OnabotulinumtoxinA:


Use OnabotulinumtoxinA as directed by your doctor. Check the label on the medicine for exact dosing instructions.


  • OnabotulinumtoxinA comes with an extra patient information sheet called a Medication Guide. Read it carefully. Read it again each time you get OnabotulinumtoxinA refilled.

  • OnabotulinumtoxinA is usually given as an injection at your doctor's office, hospital, or clinic.

  • If you miss a dose of OnabotulinumtoxinA, contact your doctor right away.

Ask your health care provider any questions you may have about how to use OnabotulinumtoxinA.



Important safety information:


  • OnabotulinumtoxinA may cause dizziness, drowsiness, loss of strength, weakness, blurred vision, or drooping eyelids. These effects may be worse if you take it with alcohol or certain medicines. Use OnabotulinumtoxinA with caution. Do not drive or perform other possibly unsafe tasks until you know how you react to it.

  • Do NOT use more than the recommended dose or use OnabotulinumtoxinA more often than every 3 months without checking with your doctor.

  • Tell your doctor or dentist that you take OnabotulinumtoxinA before you receive any medical or dental care, emergency care, or surgery.

  • Do not switch brands of OnabotulinumtoxinA. Contact your doctor if you have any questions.

  • OnabotulinumtoxinA contains albumin, which comes from human blood. There is a very rare risk of getting a viral disease or a central nervous system disease called Creutzfeldt-Jakob disease from products with albumin. No cases of these problems have been found in patients who have used OnabotulinumtoxinA.

  • OnabotulinumtoxinA should not be used in CHILDREN younger than 18 years old; safety and effectiveness in these children have not been confirmed.

  • PREGNANCY and BREAST-FEEDING: Do not use OnabotulinumtoxinA if you are pregnant. If you think you may be pregnant, contact your doctor. You will need to discuss the benefits and risks of using OnabotulinumtoxinA while you are pregnant. It is not known if OnabotulinumtoxinA is found in breast milk. If you are or will be breast-feeding while you use OnabotulinumtoxinA, check with your doctor. Discuss any possible risks to your baby.


Possible side effects of OnabotulinumtoxinA:


All medicines may cause side effects, but many people have no, or minor, side effects. Check with your doctor if any of these most COMMON side effects persist or become bothersome:



Anxiety; back pain; dizziness; drowsiness; dry mouth; dry or irritated eyes; facial pain; flu-like symptoms; headache; inability to focus eyes; increased cough; indigestion; nausea; neck pain; pain, redness, swelling, or tenderness at the injection site; runny nose; sensitivity to light; stiff or weak muscles at or near the injection site; sweating.



Seek medical attention right away if any of these SEVERE side effects occur:

Severe allergic reactions (rash; hives; itching; difficulty breathing; tightness in the chest; swelling of the mouth, face, lips, or tongue; unusual hoarseness); bleeding at the injection site; chest pain; difficulty swallowing or breathing; double or blurred vision or other vision changes; drooping of the upper eyelid; eyelid swelling; fainting; fever, chills, or persistent sore throat; irregular heartbeat; loss of bladder control; loss of strength; paralysis; seizures; severe or persistent muscle weakness or dizziness; shortness of breath; speech changes or problems.

Uses For diphtheria antitoxin

Diphtheria antitoxin is used to prevent and/or treat diphtheria infection in persons exposed to the disease.


Although rare in the U.S., diphtheria is a serious disease that can cause life-threatening illnesses. Diphtheria is transmitted through contact with an infected person or a carrier of the disease.


Diphtheria antitoxin is to be administered only by or under the supervision of your doctor or other health care professional.


Before Using diphtheria antitoxin


In deciding to use a medicine, the risks of taking the medicine must be weighed against the good it will do. This is a decision you and your doctor will make. For diphtheria antitoxin, the following should be considered:


Allergies


Tell your doctor if you have ever had any unusual or allergic reaction to diphtheria antitoxin or any other medicines. Also tell your health care professional if you have any other types of allergies, such as to foods, dyes, preservatives, or animals. For non-prescription products, read the label or package ingredients carefully.


Pediatric


Although there is no specific information comparing use of diphtheria antitoxin in children with use in other age groups, diphtheria antitoxin is not expected to cause different side effects or problems in children than it does in adults.


Geriatric


Many medicines have not been studied specifically in older people. Therefore, it may not be known whether they work exactly the same way they do in younger adults or if they cause different side effects or problems in older people. There is no specific information comparing use of diphtheria antitoxin in the elderly with use in other age groups. However, there is no evidence that the effects of diphtheria antitoxin in older adults differ from those in younger persons.


Breast Feeding


Studies in women suggest that this medication poses minimal risk to the infant when used during breastfeeding.


Interactions with Medicines


Although certain medicines should not be used together at all, in other cases two different medicines may be used together even if an interaction might occur. In these cases, your doctor may want to change the dose, or other precautions may be necessary. Tell your healthcare professional if you are taking any other prescription or nonprescription (over-the-counter [OTC]) medicine.


Interactions with Food/Tobacco/Alcohol


Certain medicines should not be used at or around the time of eating food or eating certain types of food since interactions may occur. Using alcohol or tobacco with certain medicines may also cause interactions to occur. Discuss with your healthcare professional the use of your medicine with food, alcohol, or tobacco.


Other Medical Problems


The presence of other medical problems may affect the use of diphtheria antitoxin. Make sure you tell your doctor if you have any other medical problems.


Proper Use of diphtheria antitoxin


Dosing


The dose of diphtheria antitoxin will be different for different patients. Follow your doctor's orders or the directions on the label. The following information includes only the average doses of diphtheria antitoxin. If your dose is different, do not change it unless your doctor tells you to do so.


The amount of medicine that you take depends on the strength of the medicine. Also, the number of doses you take each day, the time allowed between doses, and the length of time you take the medicine depend on the medical problem for which you are using the medicine.


  • For injection dosage form:
    • For prevention of diphtheria:
      • Adults and children—One dose injected into a muscle.


    • For treatment of diphtheria:
      • Adults and children—One to six doses injected into a muscle.

Along with its needed effects, a medicine may cause some unwanted effects. Although not all of these side effects may occur, if they do occur they may need medical attention.


Check with your doctor immediately if any of the following side effects occur:


More common
  • Chilly sensation

  • difficulty in breathing and swallowing

  • feeling of discomfort

  • fever

  • inflammation of joints

  • hives

  • itching, especially of hands or feet

  • muscle aches

  • rapid rise in body temperature

  • rash

  • reddening of skin, especially around ears

  • swelling of eyes, face, or inside of nose

  • swollen lymph glands

  • unusual tiredness or weakness, sudden and severe

Before Using benzphetamine


In deciding to use a medicine, the risks of taking the medicine must be weighed against the good it will do. This is a decision you and your doctor will make. For benzphetamine, the following should be considered:


Allergies


Tell your doctor if you have ever had any unusual or allergic reaction to benzphetamine or any other medicines. Also tell your health care professional if you have any other types of allergies, such as to foods, dyes, preservatives, or animals. For non-prescription products, read the label or package ingredients carefully.


Pediatric


Appropriate studies have not been performed on the relationship of age to the effects of benzphetamine in children below 12 years of age. Safety and efficacy have not been established .


Geriatric


Appropriate studies performed to date have not demonstrated geriatrics-specific problems that would limit the usefulness of benzphetamine in the elderly. However, elderly patients are more likely to have age-related liver, kidney, or heart problems, which may require an adjustment in the dose for patients receiving benzphetamine .


Pregnancy

All TrimestersXStudies in animals or pregnant women have demonstrated positive evidence of fetal abnormalities. This drug should not be used in women who are or may become pregnant because the risk clearly outweighs any possible benefit.

Breast Feeding


There are no adequate studies in women for determining infant risk when using this medication during breastfeeding. Weigh the potential benefits against the potential risks before taking this medication while breastfeeding.


Interactions with Medicines


Although certain medicines should not be used together at all, in other cases two different medicines may be used together even if an interaction might occur. In these cases, your doctor may want to change the dose, or other precautions may be necessary. When you are taking benzphetamine, it is especially important that your healthcare professional know if you are taking any of the medicines listed below. The following interactions have been selected on the basis of their potential significance and are not necessarily all-inclusive.


Using benzphetamine with any of the following medicines is not recommended. Your doctor may decide not to treat you with this medication or change some of the other medicines you take.


  • Brofaromine

  • Clorgyline

  • Furazolidone

  • Iproniazid

  • Isocarboxazid

  • Lazabemide

  • Linezolid

  • Moclobemide

  • Nialamide

  • Pargyline

  • Phenelzine

  • Procarbazine

  • Rasagiline

  • Selegiline

  • Toloxatone

  • Tranylcypromine

Interactions with Food/Tobacco/Alcohol


Certain medicines should not be used at or around the time of eating food or eating certain types of food since interactions may occur. Using alcohol or tobacco with certain medicines may also cause interactions to occur. Discuss with your healthcare professional the use of your medicine with food, alcohol, or tobacco.


Other Medical Problems


The presence of other medical problems may affect the use of benzphetamine. Make sure you tell your doctor if you have any other medical problems, especially:


  • Agitation or anxiety, severe or

  • Drug abuse or dependence, history of or

  • Glaucoma or

  • Heart or blood vessel disease or

  • High blood pressure, moderate or severe or

  • Overactive thyroid—benzphetamine should not be used if any of these conditions exist. Serious unwanted effects could occur .

  • Diabetes mellitus or

  • High blood pressure, mild or

  • Mental illness—Use with caution. benzphetamine may make these conditions worse .

Proper Use of benzphetamine


It is best to take benzphetamine in the mid-morning or mid-afternoon. Taking benzphetamine late in the afternoon or evening could make it harder for you to fall asleep .


Carefully follow your doctor's instructions about a special diet plan that will help you lose weight .


Dosing


The dose of benzphetamine will be different for different patients. Follow your doctor's orders or the directions on the label. The following information includes only the average doses of benzphetamine. If your dose is different, do not change it unless your doctor tells you to do so.


The amount of medicine that you take depends on the strength of the medicine. Also, the number of doses you take each day, the time allowed between doses, and the length of time you take the medicine depend on the medical problem for which you are using the medicine.


  • For oral dosage form (tablets):
    • For weight loss:
      • Adults and teenagers—At first, 25 to 50 milligrams (mg) once a day. Your doctor may increase your dose if needed. However, the dose is usually not more than 25 to 50 mg three times a day.

      • Children below 12 years of age—Use and dose must be determined by your doctor .



Missed Dose


If you miss a dose of benzphetamine, take it as soon as possible. However, if it is almost time for your next dose, skip the missed dose and go back to your regular dosing schedule. Do not double doses.


Storage


Store the medicine in a closed container at room temperature, away from heat, moisture, and direct light. Keep from freezing.


Keep out of the reach of children.


Do not keep outdated medicine or medicine no longer needed.


Ask your healthcare professional how you should dispose of any medicine you do not use.


Precautions While Using benzphetamine


It is very important that your doctor check your progress at regular visits to make sure that benzphetamine is working properly .


benzphetamine may be habit-forming. If you feel that the medicine is not working as well, do not use more than your prescribed dose. Call your doctor for instructions .


If you will be taking benzphetamine for a long time, do not stop taking it without first checking with your doctor. Your doctor may want you to gradually reduce the amount you are taking before stopping completely .


Check with your doctor right away if you think that you may be pregnant. Benzphetamine may cause birth defects or other problems in the baby if taken during pregnancy. Use an effective form of birth control to keep from getting pregnant .


You should not use benzphetamine if you have used an MAO inhibitor (MAOI) such as Eldepryl®, Marplan®, Nardil®, or Parnate® within the past 14 days .


Do not take other medicines unless they have been discussed with your doctor. This includes prescription or nonprescription (over-the-counter [OTC]) medicines, and herbal or vitamin supplements .


benzphetamine may cause some people to feel a false sense of well-being or to become dizzy, lightheaded, or less alert than they are normally. Make sure you know how you react to benzphetamine before you drive, use machines, or do anything else that could be dangerous if you are dizzy or not alert.


benzphetamine Side Effects


Along with its needed effects, a medicine may cause some unwanted effects. Although not all of these side effects may occur, if they do occur they may need medical attention.


Check with your doctor immediately if any of the following side effects occur:


More common
  • Agitation

  • anxiety

  • confusion

  • convulsions (seizures)

  • dizziness

  • fast, irregular, pounding, or racing heartbeat or pulse

  • feeling lightheaded or faint

  • hallucinations

  • headache

  • hives or welts

  • irritability

  • itching

  • lightheadedness

  • mood or other mental changes

  • nervousness

  • nightmares

  • numbness or tingling in the arms or legs

  • redness of skin

  • restlessness

  • shakiness in the legs, arms, hands, or feet

  • skin rash

  • sleeplessness

  • sweating

  • trembling of the hands or feet

  • trouble with sleeping

  • trouble with thinking, speaking, or walking

  • unable to sleep

  • unusual feeling of excitement

  • weakness

Rare
  • Chest discomfort or pain

  • difficulty breathing

  • discouragement

  • feeling sad or empty

  • lack of appetite

  • loss of interest or pleasure

  • severe mental changes

  • shortness of breath

  • swelling of the feet or lower legs

  • trouble with concentrating

  • trouble with breathing

  • unusual tiredness or weakness

Get emergency help immediately if any of the following symptoms of overdose occur:


Symptoms of overdose
  • Blurred vision

  • change in consciousness

  • dark-colored urine

  • diarrhea

  • dizziness, faintness, or lightheadedness when getting up from a lying or sitting position suddenly

  • fever

  • loss of consciousness

  • muscle cramps or spasms

  • muscle pain or stiffness

  • nausea

  • nervousness

  • physical attempt to injure oneself

  • pounding in the ears

  • rapid, shallow breathing

  • severe anxiety or feeling of panic

  • stomach cramps

  • violent actions

  • vomiting

Some side effects may occur that usually do not need medical attention. These side effects may go away during treatment as your body adjusts to the medicine. Also, your health care professional may be able to tell you about ways to prevent or reduce some of these side effects. Check with your health care professional if any of the following side effects continue or are bothersome or if you have any questions about them:


More common
  • Decrease in sexual ability, desire, drive, or performance

  • decreased interest in sexual intercourse

  • dry mouth

  • inability to have or keep an erection

  • increase in sexual ability, desire, drive, or performance

  • increased interest in sexual intercourse

  • unpleasant taste

Other side effects not listed may also occur in some patients. If you notice any other effects, check with your healthcare professional.
